(+,-)-exo,exo-2,3-Camphanediol is utilized as a conditioning agent, affording a soft texture and feel to hair and skin. It is also known to increase microcirculation, leading to its use in several products in the cosmetics industry. Moreover, it has found application as a component in protective surface coatings.
The most common route to (+,-)-exo,exo-2,3-camphanediol (Isomeric purity: >94%) is via the reduction of camphorquinone with hydride reagents such as LiAlH4 and NaBH4.[i],[ii] However, LiAlH4 is a toxic and pyrophoric reducing agent with poor solubility in common ether solvents and requires the use of strictly anhydrous reaction conditions. On the other hand, the milder NaBH4 provides the diol product in very low yields.[iii] The alternate synthesis via catalytic hydrogenation over Raney Ni necessitates the use of high pressures (~1000 psi of H2) and provides the (+,-)-exo,exo-2,3-camphanediol in poor diastereoselectivities.1
At Chemhub, we have developed a novel, mild reduction of camphorquinone to (+,-)-exo,exo-2,3-camphanediol in high yields and in excellent isomeric purity (>97%). The scalability and robustness of our method is particularly attractive, providing multi-kilogram quantities of (+,-)-exo,exo-2,3-camphanediol.
